Trichohyalin is a hair follicle protein with characteristics similar to those of profilaggrin (135940) involucrin (147360) and loricrin (152445) Trichohyalin is incorporated into the IF (intermediate filament)-like structures in the mature cells of the hair follicles inner root sheath Like profilaggrin involucrin and loricrin trichohyalin becomes highly crosslinked by the enzyme transglutaminase The trichohyalin molecule has a mass of approximately 200 kD and consists predominantly of tandem repeats of an approximately 23-amino acid sequence Fietz et al (1992) used a sheep trichohyalin cDNA probe in tritium-labeled in situ hybridization to demonstrate that the human THL gene is located in the region 1q211-q23 probably 1q213 Profilaggrin involucrin and loricrin map to the same area suggesting that the 4 proteins form a novel superfamily of epithelial structural proteins By analysis of human-rodent somatic cell hybrids and fluorescence in situ hybridization Lee et al (1993) confirmed the mapping of THL to 1q21
